Study of catalytic decomposition of silica

This thesis deals with catalytic decomposition of silica using electron microscopy and Auger spectroscopy. The thesis is primarily focused on the Au-Si phase, which is created during in the reaction. In the theoretical part, the principle of used methods is described and recherche on the catalytic decomposition of silica is presented. The practical part contains the results of the measurements and their interpretation.

There are specified requirements for a system for SAM (Software Asset Management) and also the design of the system. It deals with the description of the system implementation with the chosen implementation environment, followed with testing of this system and evaluation of results.
Study of CO oxidation on platinum by Auger spectroscopy

In the presented bachelor thesis, I deal with the study of the oxidation of carbon monoxide on platinum using Auger spectroscopy. In the introduction, I will describe the used methods. Furthermore, I theoretically describe the investigated reaction and indicate the measurements for a better understanding of the reaction. Then I will establish an experimental part, where I deal with the growth of graphene structures. This will be followed up by performing in-situ observations of the reaction with and without the presence of graphene. These measurements I compare and then analyse the surface composition during the reaction. Finally, I will create a methodology that can distinguish the individual molecules on the surface of the catalyst.

The thesis deals with the development of complex modular ultra-high vacuum system based on ultra-high vacuum scanning electron microscope UHV SEM. This discourse focuses primarily on engineering design of the system, which is underlying demands of ultra-high vacuum, limitation of mechanical vibrations and future R&D applications. The device is designed for preparing, observing and modifying of nanostructures in situ.
Using of the attack "Pass the hash attack" for the compromising of high privileged accounts.

The master thesis deals with the attack "‘pass the hash"’ on high privileged accounts. Within the theoretical part is discussed creating hashes and its use. Next is a descrip- tion of the authentication in Windows operating system. There are also pointed out weaknesses in the design of authentication mechanisms. The last part deals with the individual attack and security options for mitigating the impacts. In the practical part are tested available tools for retrieving hashes from the files of the operating systems and tools which allow the attack itself. The output of this section is selection of the appropriate tools to demonstrate the attack in a proposed real environ- ment. The last topic is about designing the experimental environment, demostration of the attack with the possibility of getting through the network. The last steps deal with mitigating the impact of the attack.
Web Application for Managing Software Detection Rules in Software Asset Management

The content of this thesis is the management of a library of rules used for software recognition on devices. The application has special purpose in the Software Asset Management system and other components depend on it. Software product recognition is performed by applying rules to the information present about the installation of the programs. Formats for writing rules and methodology may be different, but they are based on the original Alvao best practice. The work is primarily focused on simplifying and automating the process of creating or adjusting rules by means of appropriate suggesting of the resulting rules, categorization or other related actions.

Structural and functional characterization of a flaviviral methyltransferase

Recently, non-cellular viral agents became the focus of a large number of scientific groups. A prominent and widespread group of these viruses are flaviviruses, which include, for example, Zika virus, Dengue fever virus, tick-borne encephalitis virus and West Nile virus. There is a considerable diversity among these viruses, however, highly conserved proteins can be found throughout this viral genus. The largest and most conserved protein encoded by flaviviruses is the nonstructural NS5 protein. Its N-terminal domain bears the methyltransferase (MTase) activity. Thanks to the methylation of its genome, it allows the virus to initiate translation and at the same time mask it from the host's immune system. By blocking the active site of this enzyme with a small molecule, viral infection could be stopped not only in one flavivirus, but, due to the high conservation of MTases, in all other flaviviruses. This diploma thesis deals with the aforementioned MTase domain of the NS5 protein, specifically of the West Nile virus (WNV). After designing an insert encoding the WNV MTase domain, amplifying it and ligating it into the vector, the MTase domain was prepared by a recombinant expression, followed by purification.
